Characterization of Polyphenol Oxidase in Subceflular Fraction from Ginkgo biloba L. Leaves

Abstract
We examined the effect of pH on the polyphenoloxidase (PPO) activities of 200x g(cell wall), 4,000xg(plastid), 100,000xg(mitochondiia) and soluble fractions of Ginkgo biloba L. leaves. Each subcellular fraction showed similar optimum pHs. The substrate specificity of enzyme activities was also investigated at optimum pH. All fractions were highly specific to 4-methylcatechol and gallic acid and the soluble fraction was specific to hydrocaffeic acid. And other substrates tested in this study showed different specificities.
